Design Notes
The sequence for the P32 promoter and RBS was obtained from Zhu et al. (2015) in their paper "Isolation of strong constitutive promoters from Lactococcus lactis subsp. Lactis N8." Our lab uses the Lee/Dueber YTK Golden Gate standard, which, in terms of part overhangs, is different than the iGEM PhytoBrick standard. Thus, we had to redesign our P32 promoter sequence to contain the proper overhangs specified by the iGEM PhytoBrick standard.
Source
The P32 promoter and its RBS sequence are natively found in the genome of Lactococcus lactis. Using the sequence information provided by Zhu et al. (2015), we ordered the promoter and RBS sequence as a gBlock from IDT.
